Description

The MFHAS1 (multifunctional ROCO family signaling regulator 1) is a protein-coding gene located on chromosome 8.

MFHAS1 is a protein that likely binds GTP. It plays a role in innate immunity, particularly in the inflammatory response, by regulating the Toll-like receptor (TLR) signaling pathways TLR2 and TLR4. It negatively regulates the TLR4 signaling pathway that activates the transcription factor AP-1, by retaining the phosphatase complex PP2A in the cytoplasm. This prevents the dephosphorylation of the AP-1 subunit JUN, which is essential for the transcription factor's activation. MFHAS1 also exhibits a dual role in the TLR2 signaling pathway, both inhibiting and activating it. It specifically activates the downstream p38 and JNK MAP kinases within the TLR2 pathway, promoting macrophage polarization towards the pro-inflammatory M1 phenotype. Additionally, MFHAS1 might regulate inflammation induced by high glucose through the PKB/AKT signaling pathway. It has also been implicated in erythrocyte differentiation by activating the ERK1/ERK2 signaling pathway.
