Description

The MAPK6 (mitogen-activated protein kinase 6) is a protein-coding gene located on chromosome 15.

MAPK6 is an enzyme encoded by the MAPK6 gene in humans. It belongs to the Ser/Thr protein kinase family and is closely related to mitogen-activated protein kinases (MAP kinases) or extracellular signal-regulated kinases (ERKs). MAP kinases are activated through protein phosphorylation cascades and act as integration points for multiple biochemical signals. MAPK6 is located in the nucleus and is activated in fibroblasts upon treatment with serum or phorbol esters. It was initially cloned from the rat brain cDNA library using a probe derived from ERK1. The MAPK6 gene is located on chromosome 15 (15q21.2) and spans 47.01 kb. It consists of 6 exons, with the translation initiation codon located in exon 2. MAPK6 is an atypical member of the MAP kinase family with a molecular mass of approximately 100 kDa and is composed of 721 amino acid residues.

MAPK6 is an atypical mitogen-activated protein kinase (MAPK) that phosphorylates microtubule-associated protein 2 (MAP2) and MAPKAPK5. The role of the complex formed with MAPKAPK5 is not fully understood, but it involves a series of phosphorylation events: MAPKAPK5 phosphorylates MAPK6 at Ser-189, which in turn activates MAPKAPK5. MAPKAPK5 then phosphorylates MAPK6. This complex may also play a role in promoting cell cycle entry.
