MAML1


Description

The MAML1 (mastermind like transcriptional coactivator 1) is a protein-coding gene located on chromosome 5.

MAML1, the human homolog of mastermind, a Drosophila protein involved in the Notch signaling pathway, is a transcriptional coactivator in this pathway. It forms a complex with the intracellular portion of human Notch receptors and can increase expression of a Notch-induced gene. MAML1 interacts with EP300 and NOTCH1.

MAML1 acts as a transcriptional coactivator for NOTCH proteins, amplifying NOTCH-induced transcription of HES1. It enhances phosphorylation and proteolytic turnover of the NOTCH intracellular domain in the nucleus through interaction with CDK8. MAML1 binds to CREBBP/CBP, promoting nucleosome acetylation at NOTCH enhancers and activating transcription. It induces phosphorylation and localization of CREBBP to nuclear foci. MAML1 plays a role in hematopoietic development by regulating NOTCH-mediated lymphoid cell fate decisions.
