MACROD1


Description

The MACROD1 (mono-ADP ribosylhydrolase 1) is a protein-coding gene located on chromosome 11.

MACROD1, also known as MACRO domain-containing protein 1, is an ADP-ribose glycohydrolase that removes ADP-ribose from proteins. It plays a role in estrogen signaling and may be involved in cancer development.

MACROD1 removes ADP-ribose from single ADP-ribose moieties attached to aspartate and glutamate residues in proteins, but it does not affect proteins with poly-ADP-ribose. It also deacetylates O-acetyl-ADP ribose, a signaling molecule generated by the deacetylation of acetylated lysine residues in histones and other proteins. MACROD1 is involved in estrogen signaling and binds to the androgen receptor (AR), amplifying its transactivation function in response to androgen. It may play a role in the progression of hormone-dependent cancers by activating ESR1 transactivation. MACROD1 could act as an ESR1 coactivator, providing a positive feedback loop for ESR1 signal transduction. It may also be involved in invasive growth by downregulating CDH1 in endometrial cancer cells and enhances ESR1-mediated transcription activity.

MACROD1 is also known as LRP16.
