LRWD1


Description

The LRWD1 (leucine rich repeats and WD repeat domain containing 1) is a protein-coding gene located on chromosome 7.

LRWD1 is essential for the transition from the G1 to S phase of the cell cycle. During G1, it recruits and stabilizes the origin recognition complex (ORC) to chromatin, establishing the pre-replication complex (preRC). LRWD1 also localizes to heterochromatic sites in post-replicated cells. LRWD1 binds to both DNA and histone methylation repressive marks on heterochromatin, specifically recognizing histone H3 and H4 trimethylation marks (H3K9me3, H3K27me3, and H4K20me3) in a cooperative manner with DNA methylation. This function is required for the silencing of major satellite repeats. LRWD1 may play a role in stabilizing ORC2, ORC3, and ORC4.
