Description

The KCTD6 (potassium channel tetramerization domain containing 6) is a protein-coding gene located on chromosome 3.

KCTD6, also known as KCASH3, is a protein involved in ubiquitination and protein degradation. It acts as a substrate-specific adapter in a complex that tags proteins for degradation, potentially playing a role in regulating cell growth and development.

KCTD6 acts as a substrate-specific adapter within the BCR (BTB-CUL3-RBX1) E3 ubiquitin-protein ligase complex. This complex facilitates the ubiquitination and subsequent degradation of target proteins through the proteasome. Notably, KCTD6 promotes the ubiquitination of HDAC1, a process that appears to rely on the oligomerization of KCTD11 and KCTD6. KCTD6 can also function as an antagonist of the Hedgehog pathway by influencing the nuclear translocation of the transcription factor GLI1. This antagonism is likely mediated by HDAC1 down-regulation, which keeps GLI1 acetylated and inactive. Furthermore, KCTD6 has been shown to inhibit cell growth and tumorigenicity in medulloblastoma. It also plays a role in regulating the protein levels of the ANK1 isoform Mu17, likely through CUL3-dependent proteasomal degradation.
