Description

The ITPKB (inositol-trisphosphate 3-kinase B) is a protein-coding gene located on chromosome 1.

ITPKB is an enzyme encoded by the ITPKB gene in humans. It is one of three isoforms of Inositol-trisphosphate 3-kinase found in humans. ITPKB regulates inositol phosphate metabolism by phosphorylating the second messenger InsP3, which triggers calcium release from intracellular stores in the endoplasmic reticulum via the inositol trisphosphate receptor. However, ITPKB produces Ins(1,3,4,5)P4, which does not activate the inositol trisphosphate receptor. The enzyme specifically phosphorylates the 1,4,5 isomer of IP3. ITPKB's activity controls the levels of various inositol polyphosphates crucial for cellular signaling. Its activity is regulated by both calcium/calmodulin and protein phosphorylation mechanisms. ITPKB plays a vital role in immune cell function and is essential for T and B cell development.

ITPKB catalyzes the phosphorylation of InsP3 (inositol 1,4,5-trisphosphate) to produce Ins(1,3,4,5)P4 (inositol 1,3,4,5-tetrakisphosphate). This enzymatic activity plays a role in regulating calcium homeostasis within cells.
