INPP4B (Inositol polyphosphate-4-phosphatase, type II, 105kDa) is a protein encoded by the INPP4B gene in humans. It is a dual specificity phosphatase that plays a role in phosphatidylinositol signaling pathways. INPP4B removes the phosphate group at position 4 of the inositol ring from inositol 3,4-bisphosphate and phosphate groups from phosphotyrosines. Limited evidence suggests that the human type II enzyme undergoes alternative splicing, unlike the type I enzyme.

INPP4B removes phosphate groups from specific positions on inositol phosphates, including phosphatidylinositol 3,4-bisphosphate (PIP2), inositol 1,3,4-trisphosphate (IP3), and inositol 3,4-trisphosphate (IP3). This activity is crucial for its role in regulating cell processes, such as macropinocytosis. It also inhibits the PI3K-AKT/PKB signaling pathway, a pathway associated with cell growth and survival, by removing phosphate groups from phosphoinositides. This function is thought to contribute to INPP4B's tumor suppressor activity.
