Description

The ING4 (inhibitor of growth family member 4) is a protein-coding gene located on chromosome 12.

ING4 is a protein encoded by the ING4 gene. It is similar to ING1, a tumor suppressor protein that interacts with TP53, inhibits cell growth, and induces apoptosis. ING4 contains a PHD-finger, a common motif in chromatin remodeling proteins. It binds TP53 and EP300/p300, a component of the histone acetyl transferase complex, suggesting its role in the TP53-dependent regulatory pathway. ING4 interacts with EP300, RELA, and P53.

ING4 is a component of HBO1 complexes, which specifically acetylate histone H3 at Lys-14 (H3K14ac) and have reduced activity toward histone H4. This acetylation may play a role in DNA replication. ING4 can inhibit tumor progression by regulating the transcriptional output of cell proliferation pathways. It can suppress brain tumor angiogenesis by repressing RELA/NFKB3 target genes when complexed with RELA. ING4 may also suppress loss of contact inhibition induced by oncogenes like MYC. It represses hypoxia inducible factor (HIF) activity by interacting with HIF prolyl hydroxylase 2 (EGLN1). ING4 can enhance apoptosis induced by serum starvation in mammary epithelial cells.
