The IL22 (interleukin 22) is a protein-coding gene located on chromosome 12.

IL-22 is a cytokine that plays a key role in regulating tissue responses during inflammation. It is essential for epithelial cell regeneration, maintaining barrier function after injury, and preventing further damage. Unlike most cytokines, it doesn't affect immune cells. IL-22 binds to a heterodimeric receptor composed of IL22RA1 and IL10RB. IL22RA1 is found on non-immune cells in various organs. Binding activates JAK1 and TYK2 tyrosine kinases, which then activate STAT3. STAT3, ERK1/2, and PI3K/AKT pathways promote cell survival and proliferation. IL-22 also promotes phosphorylation of GSK3B at Ser-9 and CTTN, leading to epithelial cell spreading.
