IL1R1
Description
The IL1R1 (interleukin 1 receptor type 1) is a protein-coding gene located on chromosome 2.
IL1R1, also known as CD121a, is a cytokine receptor belonging to the interleukin-1 receptor family. It binds to interleukin-1 alpha (IL1A), interleukin 1 beta (IL1B), and interleukin 1 receptor antagonist (IL1RA). This receptor is a crucial mediator involved in many immune and inflammatory responses triggered by cytokines. IL1R1 is located on chromosome 2q12, within a gene cluster that also includes IL1R2, IL1RL2, and IL1RL1. IL1R1 interacts with PIK3R1, Myd88, and IL1RAP.
IL1R1 binds to interleukin-1 (IL-1) family members, including IL1A, IL1B, and IL1RN, with comparable affinity. Upon binding to IL-1, IL1R1 associates with the coreceptor IL1RAP, forming a high-affinity complex. This complex activates signaling pathways, including NF-kappa-B, MAPK, and others, through the recruitment of adapter molecules such as TOLLIP, MYD88, and IRAK1 or IRAK2. The antagonist IL1RN prevents the formation of the signaling complex by blocking the association with IL1RAP. IL1R1 plays a role in IL1B-mediated costimulation of IFNG production from T-helper 1 (Th1) cells.
