Description

The HOMER3 (homer scaffold protein 3) is a protein-coding gene located on chromosome 19.

HOMER3 is a protein encoded by the HOMER3 gene in humans. It belongs to the homer family of dendritic proteins, which regulate the function of group 1 metabotrophic glutamate receptors. HOMER3 may be involved in cell growth. It interacts with TRPC1 and RYR1.

HOMER3 acts as a scaffolding protein in the postsynaptic density, linking together various proteins involved in neuronal signaling. It binds to and cross-links cytoplasmic regions of receptors like GRM1, GRM5, ITPR1, and others, facilitating the connection between surface receptors and intracellular calcium release. Different isoforms of HOMER3 can be regulated in unique ways, potentially playing a critical role in maintaining the flexibility of glutamatergic synapses. Furthermore, HOMER3 inhibits the activation of T cells by interfering with the calcineurin-NFAT pathway. This inhibition occurs through competition with calcineurin for binding to the NFAT protein, preventing NFAT activation.

HOMER3 is also known as HOMER-3, VESL3.
