HOMER2
Description
The HOMER2 (homer scaffold protein 2) is a protein-coding gene located on chromosome 15.
HOMER2, also known as Homer protein homolog 2, is a protein encoded by the HOMER2 gene in humans. It belongs to the homer family of dendritic proteins that regulate group 1 metabotrophic glutamate receptor function. HOMER2 may play a role in cell growth. Four transcript variants encoding distinct isoforms have been identified for this gene. HOMER2 has been shown to interact with RYR1.
HOMER2, a postsynaptic density scaffolding protein, plays a crucial role in linking surface receptors to intracellular calcium release. It achieves this by physically connecting GRM1 and GRM5 with ER-associated ITPR1 receptors. Additionally, HOMER2 might link GRM1 to PI3 kinase through its interaction with AGAP2. Different isoforms of HOMER2 exhibit distinct regulatory mechanisms and contribute to the maintenance of plasticity at glutamatergic synapses. HOMER2 is essential for normal hearing and acts as a negative regulator of T cell activation. This regulation is achieved by inhibiting the calcineurin-NFAT pathway through competition with calcineurin/PPP3CA for binding to NFAT proteins. This interaction ultimately prevents NFAT activation by PPP3CA.
