HLTF
Description
The HLTF (helicase like transcription factor) is a protein-coding gene located on chromosome 3.
HLTF is an enzyme encoded by the HLTF gene in humans. It belongs to the SWI/SNF family, which possesses helicase and ATPase activities and is thought to regulate gene transcription by modifying chromatin structure. HLTF contains a RING finger DNA binding motif, and two transcript variants have been identified, both encoding the same protein. However, an alternative translation start site produces an isoform truncated at the N-terminus compared to the full-length protein. HLTF is a double-stranded DNA translocase, one of two human homologs of Saccharomyces cerevisiae RAD5, alongside SHPRH (SNF2 histone linker PHD RING helicase), capable of performing fork regression, similar to Rad5. HLTF has been shown to interact with UBE2N, RAD18, and UBE2V2.
HLTF possesses both helicase and E3 ubiquitin ligase activities. It has intrinsic ATP-dependent nucleosome-remodeling activity, which may be necessary for the activation or repression of specific target promoters. This includes the SERPINE1 and HIV-1 promoters, and the SV40 enhancer, to which HLTF can bind directly. HLTF plays a role in error-free postreplication repair (PRR) of damaged DNA and maintains genomic stability by acting as a ubiquitin ligase for 'Lys-63'-linked polyubiquitination of chromatin-bound PCNA.
