Description

The GSPT1 (G1 to S phase transition 1) is a protein-coding gene located on chromosome 16.

The GSPT1 gene in humans encodes the eukaryotic peptide chain release factor GTP-binding subunit ERF3A, an enzyme involved in translation termination. GSPT1 interacts with BIRC2.

GSPT1, also known as ERF3A, is a GTPase that acts as a component of the eRF1-eRF3-GTP ternary complex, which mediates translation termination in response to the stop codons UAA, UAG, and UGA. GSPT1 delivers ETF1/ERF1 to stop codons by binding to the ribosomal A-site. GTP hydrolysis by GSPT1 induces a conformational change that leads to its dissociation, allowing ETF1/ERF1 to fully occupy the A-site. GSPT1 is also a component of the transient SURF complex, which recruits UPF1 to stalled ribosomes during nonsense-mediated decay (NMD) of mRNAs containing premature stop codons. GSPT1 is required for SHFL-mediated translation termination, which inhibits programmed ribosomal frameshifting (-1PRF) of mRNA from viruses and cellular genes.
