The GRB14 (growth factor receptor bound protein 14) is a protein-coding gene located on chromosome 2.

GRB14, or Growth factor receptor-bound protein 14, is a protein encoded by the GRB14 gene in humans. It belongs to a small family of adapter proteins that interact with various receptor tyrosine kinases and signaling molecules. GRB14 binds to insulin receptors and insulin-like growth-factor receptors, likely having an inhibitory effect on receptor tyrosine kinase signaling, especially insulin receptor signaling. This gene may play a role in signaling pathways regulating growth and metabolism. Transcript variants have been reported for this gene, but their full-length natures have not been determined. GRB14 interacts with Epidermal growth factor receptor, Fibroblast growth factor receptor 1 and TNKS2.

GRB14 modulates the coupling of cell surface receptor kinases to specific signaling pathways. It binds to and suppresses signals from the activated insulin receptor (INSR). GRB14 is a potent inhibitor of insulin-stimulated MAPK3 phosphorylation. It plays a crucial role in regulating PDPK1 membrane translocation in response to insulin stimulation. It acts as an adapter protein, recruiting PDPK1 to the activated insulin receptor, thereby promoting PKB/AKT1 phosphorylation and insulin signal transduction.
