The GNPDA2 (glucosamine-6-phosphate deaminase 2) is a protein-coding gene located on chromosome 4.

GNPDA2 is a human gene that encodes the enzyme Glucosamine-6-phosphate deaminase 2. This enzyme is involved in the regulation of UDP-GlcNAc synthesis, a crucial molecule in the hexosamine pathway. Variations in the GNPDA2 gene have been linked to obesity.

GNPDA2 catalyzes the interconversion of glucosamine-6-phosphate (GlcN-6P) and fructose-6-phosphate (Fru-6P), a key regulatory step in the hexosamine biosynthesis pathway. This reversible reaction involves deamination and aldo-keto isomerization, influencing the metabolic flow between UDP-GlcNAc and Fru-6P. High ammonium levels can promote the conversion of Fru-6P to UDP-GlcNAc, contributing to the regulation of UDP-GlcNAc levels in the cell. This regulation impacts hyaluronan synthesis during tissue remodeling.
