Zinc finger protein 281 is a protein that in humans is encoded by the ZNF281 gene. It is a transcription factor with zinc finger domains. These domains are known to bind DNA and play a role in gene regulation.
ZNF281 is a transcription repressor that plays a key role in regulating embryonic stem cell (ESC) differentiation. It is essential for ESC differentiation and functions by mediating the autorepression of NANOG in ESCs. This involves binding to the NANOG promoter, promoting the association of NANOG protein to its own promoter, and recruiting the NuRD complex, which deacetylates histones. While ZNF281 is not required for the establishment and maintenance of ESCs, it does repress the transcription of several genes including GAST, ODC1, and VIM. It binds to the G-rich box within the enhancer region of these genes.
