ZFYVE9 (Zinc finger FYVE domain-containing protein 9) or SARA (SMAD anchor for receptor activation) is a protein that plays a role in TGF beta signaling. It contains a double zinc finger (FYVE domain) and functions as an anchoring protein. SARA binds to the MH2 domain of the R-SMADs SMAD2 and SMAD3, as well as the type I TGF beta receptors. This binding facilitates the phosphorylation of the R-SMAD, which subsequently dissociates from SARA and the receptor and binds a coSMAD. The complex then enters the nucleus and acts as transcription factors.
ZFYVE9, also known as SARA, is an early endosomal protein that plays a crucial role in TGF-beta signaling. It facilitates the recruitment of SMAD2 and SMAD3 to intracellular membranes and the TGF-beta receptor, influencing their subcellular localization. This function regulates the transcriptional activity of the SMAD3/SMAD4 complex, potentially contributing to TGF-beta receptor internalization.
