Palmitoyltransferase ZDHHC17 is an enzyme that contains a DHHC domain that in humans is encoded by the ZDHHC17 gene.

ZDHHC17 is a palmitoyltransferase that adds palmitate to various proteins, influencing a range of cellular processes. It can also attach myristate and stearate to proteins. ZDHHC17 specifically palmitoylates neuronal proteins like SNAP25, DLG4/PSD95, GAD2, SYT1, and HTT. Additionally, it palmitoylates neuronal protein GPM6A, as well as SPRED1 and SPRED3. ZDHHC17 might also play a role in axonogenesis by regulating NTRK1 and the ERK1/ERK2 signaling cascade. It may be involved in sorting or targeting crucial proteins for endocytosis at the plasma membrane and could also contribute to Mg(2+) transport. Furthermore, ZDHHC17 can palmitoylate DNAJC5, influencing its localization to the Golgi membrane, and palmitoylates CASP6, preventing its dimerization and activation.
