The USP49 (ubiquitin specific peptidase 49) is a protein-coding gene located on chromosome 6.

USP49 specifically removes ubiquitin from histone H2B at lysine 120 (H2BK120Ub). This deubiquitination is crucial for efficient cotranscriptional splicing of a large number of exons. H2BK120Ub acts as a tag for epigenetic transcriptional activation and is involved in the regulation of mRNA splicing.
