Description

The UBE2F (ubiquitin conjugating enzyme E2 F (putative)) is a protein-coding gene located on chromosome 2.

UBE2F is a human gene that encodes a protein known as NEDD8-conjugating enzyme UBE2F. This protein plays a crucial role in the neddylation pathway, a process that involves the attachment of the ubiquitin-like protein NEDD8 to other proteins. Specifically, UBE2F accepts NEDD8 from the UBA3-NAE1 E1 complex and catalyzes its covalent attachment to other proteins. In conjunction with the E3 ubiquitin ligase RNF7/RBX2, UBE2F is responsible for neddylating cullin-5 (CUL5), a key component in the ubiquitin-proteasome system. This neddylation of CUL5 is essential for regulating the activity of the CUL5-based E3 ubiquitin ligase complex, which plays a role in various cellular processes. Notably, UBE2F does not neddylate other cullin family members, such as CUL1, CUL2, CUL3, CUL4A, or CUL4B. Additionally, UBE2F is implicated in HIV-1 infection. Following infection by HIV-1, UBE2F participates in the HIV-1 Vif protein-mediated ubiquitination and degradation of APOBEC3G by mediating the neddylation of cullin-5 (CUL5). UBE2F interacts with several proteins, including UBA3, RBX2, and various DCUN1D proteins. These interactions contribute to its role in the neddylation pathway and its involvement in cellular processes and viral infection.

UBE2F acts as a NEDD8-conjugating enzyme, receiving NEDD8 from the UBA3-NAE1 E1 complex and facilitating its attachment to other proteins. Its specific interaction with the E3 ubiquitin ligase RBX2, but not RBX1, suggests that the RBX2-UBE2F complex specifically neddylates target proteins like CUL5.
