Description

The TRAF3IP3 (TRAF3 interacting protein 3) is a protein-coding gene located on chromosome 1.

TRAF3-interacting JNK-activating modulator is a protein that in humans is encoded by the TRAF3IP3 gene.

TRAF3IP3 plays crucial roles in both innate and adaptive immunity, including thymocyte development, regulatory T-cell stability, and TLR4 signaling. It facilitates TCR-stimulated activation by recruiting MAP2K1/MEK1 to the Golgi, promoting its interaction with BRAF. TRAF3IP3 also recruits PPP2CA to the lysosome, enabling PP2Ac interaction with RPTOR and restricting glycolytic metabolism in regulatory T-cells. Additionally, it acts as a molecular clamp to facilitate LPS-induced translocation of TLR4 to lipid rafts, enhancing TLR4 signaling. During viral infection, TRAF3IP3 facilitates TRAF3 recruitment to MAVS within mitochondria, leading to IRF3 activation and interferon production. However, it also maintains immune homeostasis by promoting Lys-48-dependent ubiquitination of TBK1, preventing overzealous innate immunity.

TRAF3IP3 is also known as T3JAM.
