SPRY4
Description
The SPRY4 (sprouty RTK signaling antagonist 4) is a protein-coding gene located on chromosome 5.
SPRY4, the human gene encoding sprouty homolog 4, plays a role in inhibiting the receptor-transduced mitogen-activated protein kinase (MAPK) signaling pathway. It functions upstream of RAS activation, preventing the formation of active GTP-RAS. SPRY4 interacts with TESK1.
SPRY4 inhibits the MAPK signaling pathway triggered by insulin receptor and EGFR, but does not affect MAPK activation caused by a constitutively active mutant Ras. It potentially disrupts the formation of GTP-Ras. SPRY4 inhibits Ras-independent RAF1 activation but not Ras-dependent activation. It also prevents integrin-mediated cell spreading by inhibiting TESK1-mediated phosphorylation of cofilin.
SPRY4 is also known as HH17.
