SMYD5


Description

The SMYD5 (SMYD family member 5) is a protein-coding gene located on chromosome 2.

SMYD5 is a histone methyltransferase that specifically trimethylates lysine 20 of histone H4 (H4K20me3), leading to epigenetic transcriptional repression. It participates in the repression of TLR4-target inflammatory genes in macrophages by forming H4K20me3 at their promoters, in association with the NCoR corepressor complex. SMYD5 plays a significant role in the self-renewal and differentiation of embryonic stem cells (ESCs). It maintains ESCs by silencing differentiation genes through the deposition of H4K20me3 marks. Furthermore, it contributes to the genomic stability of ESCs during differentiation by regulating heterochromatin formation and repressing endogenous repetitive DNA elements through H4K20me3 deposition.

SMYD5 is also known as NN8-4AG, RAI15, RRG1, ZMYND23.
