SLC5A3 is a sodium-coupled sugar symporter that transports myo-inositol and its stereoisomer scyllo-inositol across the plasma membrane. It maintains myo-inositol gradients, crucial for cell volume and fluid balance, especially during osmotic stress. SLC5A3 forms coregulatory complexes with voltage-gated potassium (K+) ion channels, impacting their ion selectivity, voltage dependence, and gating kinetics. This interaction modulates myo-inositol transport, affecting neuron excitability and regulating myo-inositol gradients between blood and cerebrospinal fluid. SLC5A3 provides myo-inositol for phosphoinositide synthesis, indirectly impacting phosphoinositide-dependent ion channels and calcium signaling during osmotic stress.
