Description
The SESN3 (sestrin 3) is a protein-coding gene located on chromosome 11.
Sestrin 3 is a protein encoded by the SESN3 gene in humans. It belongs to the sestrin family of stress-induced proteins. Sestrin 3 reduces levels of intracellular reactive oxygen species (ROS) triggered by activated Ras downstream of RAC-alpha serine/threonine-protein kinase (Akt) and FoxO transcription factor. It is essential for normal blood glucose regulation, insulin resistance, and plays a role in lipid storage during obesity. Alternative splicing leads to multiple transcript variants.
Sestrin-3 functions as an intracellular leucine sensor that negatively regulates the TORC1 signaling pathway. It may also regulate the insulin-receptor signaling pathway through activation of TORC2. This metabolic regulator may play a role in protection against oxidative and genotoxic stresses by preventing the accumulation of reactive oxygen species (ROS) through the alkylhydroperoxide reductase activity of its N-terminal domain.
