Description

The RNF26 (ring finger protein 26) is a protein-coding gene located on chromosome 11.

RNF26 is a protein encoded by the RNF26 gene in humans. It contains a C3HC5 type of RING finger, a motif involved in protein-DNA and protein-protein interactions. The expression of this gene is upregulated in cancer cell lines from various types of cancer.

RNF26 is an E3 ubiquitin ligase that plays a crucial role in endosome organization. It retains vesicles in the perinuclear cloud by mediating the ubiquitination of SQSTM1, a process facilitated by its interaction with the ubiquitin conjugating enzyme UBE2J1. Ubiquitinated SQSTM1 acts as a platform for perinuclear positioning of the endosomal system by attracting vesicle-associated adapters, creating a molecular bridge that restrains cognate vesicles in the perinuclear region and optimizes cargo transport. Additionally, RNF26 regulates type I interferon production in response to viral infection. It mediates the formation of Lys-11-linked polyubiquitin chains on TMEM173/STING, stabilizing this protein and promoting interferon response. Conversely, RNF26 also limits type I interferon response by promoting the autophagic degradation of IRF3.
