Description

The RANBP3 (RAN binding protein 3) is a protein-coding gene located on chromosome 19.

Ran-binding protein 3 is a protein encoded by the RANBP3 gene in humans. This protein features a RanBD1 domain and is found in both the nucleus and cytoplasm. It plays a role in nuclear export as part of a heteromeric complex. Different isoforms of the protein are generated through alternative splicing. RanBP3 has been shown to interact with RCC1 and XPO1.

RanBP3 acts as a cofactor for XPO1/CRM1-mediated nuclear export, potentially serving as a scaffolding protein within the export complex. Its binding to XPO1/CRM1 stabilizes the interaction between XPO1/CRM1 and the cargo being transported. In the absence of Ran-bound GTP, RanBP3 prevents XPO1/CRM1 from binding to the nuclear pore complex. It binds to CHC1/RCC1, enhancing the guanine nucleotide exchange activity of CHC1/RCC1 and facilitating the Ran-dependent recruitment of XPO1/CRM1 to CHC1/RCC1. RanBP3 also plays a role in regulating TGF-beta signaling. It interacts with the R-SMAD proteins, SMAD2 and SMAD3, promoting their nuclear export and thereby acting as a negative regulator of TGF-beta signaling.
