LEMD2 is a nuclear lamina-associated inner nuclear membrane protein that plays a role in nuclear structure organization, maintenance of nuclear envelope (NE) integrity, and NE reformation after mitosis. It acts as a transmembrane adapter for the endosomal sorting complexes required for transport (ESCRT), facilitating ESCRT-mediated NE reformation. LEMD2 promotes NE closure by recruiting CHMP7 and other ESCRT-III proteins to the reforming NE during anaphase. During nuclear reassembly, it condenses into a liquid-like coating around microtubule spindles, forming a macromolecular O-ring seal to facilitate early nuclear sealing. LEMD2 is involved in the organization of heterochromatin associated with the NE and the maintenance of NE organization under mechanical stress. It is required for embryonic development, regulating signaling pathways such as MAPK and AKT. LEMD2 is also essential for myoblast differentiation and cardiac homeostasis.
