Description

The PRDX5 (peroxiredoxin 5) is a protein-coding gene located on chromosome 11.

PRDX5 (Peroxiredoxin-5, mitochondrial) is an antioxidant enzyme encoded by the PRDX5 gene on chromosome 11. It belongs to the peroxiredoxin family, which is known for its antioxidant properties. PRDX5 is widely expressed in various tissues and exhibits a unique subcellular distribution, being found in mitochondria, peroxisomes, the cytosol, and the nucleus. It is homologous to the yeast peroxisomal antioxidant enzyme PMP20. PRDX5 acts as a peroxidase that utilizes cytosolic or mitochondrial thioredoxins to reduce alkyl hydroperoxides or peroxynitrite with high rate constants. Its reaction with hydrogen peroxide is slower. PRDX5 plays a key role in protecting cells from the harmful effects of reactive oxygen species.

Structurally, PRDX5 differs from other peroxiredoxins. It is the most divergent isoform in mammals, with only 28% to 30% sequence identity to typical 2-Cys and 1-Cys peroxiredoxins. This atypical structure is reflected in its unique crystal structure. While typical peroxiredoxins have a thioredoxin domain and a C-terminal, PRDX5 has an N-terminal domain and a unique alpha helix replacing a loop structure in the thioredoxin domain.
