Description

The EXO5 (exonuclease 5) is a protein-coding gene located on chromosome 1.

EXO5 is a single-stranded DNA (ssDNA) exonuclease that plays a crucial role in DNA repair. It exhibits both 5'-3' and 3'-5' exonuclease activities, with a stronger preference for 5'-ends. EXO5 functions as a sliding exonuclease, loading at the ends of ssDNA and moving along the strand before cleaving it. However, its sliding and 3'-5' exonuclease activities are inhibited upon binding to the Replication Protein A (RPA) complex, which enforces 5'-directionality.

EXO5 is also known as C1orf176, DEM1, Exo V, hExo5.
