Description

The ADIPOR1 (adiponectin receptor 1) is a protein-coding gene located on chromosome 1.

Adiponectin receptor 1 (AdipoR1) is a protein encoded by the ADIPOR1 gene in humans. It belongs to the progestin and adipoQ receptor (PAQR) family and is also known as PAQR1. AdipoR1, similar to G protein-coupled receptors (GPCRs), has 7 transmembrane domains. However, it is oriented oppositely to GPCRs in the membrane, with a cytoplasmic N-terminus and an extracellular C-terminus. Unlike GPCRs, AdipoR1 does not associate with G proteins. AdipoR1, along with AdipoR2, functions as a receptor for both globular and full-length adiponectin. It mediates increased AMPK and PPAR-α ligand activities, promoting fatty acid oxidation and glucose uptake by adiponectin. There have been allegations of fabricated data regarding the identification of AdipoR1 and AdipoR2, which are currently under investigation. Adiponectin agonists include peptides like adiponectin, ADP-355, and ADP-399, and non-peptides like AdipoRon, (−)-Arctigenin, Arctiin, Gramine, and Matairesinol. The peptide ADP-400 acts as an antagonist.

Adiponectin receptor 1 (AdipoR1) serves as a receptor for the hormone adiponectin (ADIPOQ), which is secreted by adipocytes and plays a crucial role in regulating glucose and lipid metabolism. Adiponectin binding to AdipoR1 activates a signaling cascade, leading to increased AMP-activated protein kinase (AMPK) activity. This ultimately results in enhanced fatty acid oxidation, increased glucose uptake, and decreased gluconeogenesis. AdipoR1 has a high affinity for globular adiponectin and a lower affinity for full-length adiponectin.
