NFX1


Description

The NFX1 (nuclear transcription factor, X-box binding 1) is a protein-coding gene located on chromosome 9.

NFX1 is a protein that in humans is encoded by the NFX1 gene. It acts as a transcriptional repressor, binding to the X box motif of MHC class II genes, including HLA-DRA, and regulating their expression. This regulation is thought to be involved in controlling the duration of inflammatory responses by limiting the expression of class II MHC molecules induced by IFN-γ. Three different isoforms of NFX1, each encoded by a different splice variant, have been identified.

NFX1 binds to the X-box motif of MHC class II genes and represses their expression, potentially controlling the duration of an inflammatory response by limiting MHC class II induction by interferon-gamma. Isoform 3 of NFX1 also binds to the X-box motif of the TERT promoter, repressing its expression. In contrast, isoform 1, in conjunction with PABPC1 or PABPC4, acts as a coactivator for TERT expression. NFX1 mediates E2-dependent ubiquitination.
