The NDST3 (N-deacetylase and N-sulfotransferase 3) is a protein-coding gene located on chromosome 4.

NDST3 is a human gene encoding an enzyme that plays a crucial role in heparin production. It catalyzes the transfer of a sulfate group to glucosamine residues in heparan sulfate, a step essential for heparin biosynthesis. Mutations in NDST3 have been linked to schizophrenia.

NDST3 is a key enzyme in the production of heparin, a crucial anticoagulant. It catalyzes both the removal of an acetyl group (N-deacetylation) and the addition of a sulfate group (N-sulfation) from glucosamine residues within heparan sulfate. This modification, converting GlcNAc-GlcA to N-sulfated heparosan, is essential for subsequent steps in heparin biosynthesis. Despite its dual function, NDST3 exhibits greater deacetylase activity than sulfotransferase activity.

NDST3 is also known as HSST3.
