The MYT1L (myelin transcription factor 1 like) is a protein-coding gene located on chromosome 2.

The MYT1L gene encodes a protein belonging to the zinc finger superfamily of transcription factors, which are specialized proteins that regulate gene expression. This protein is primarily found in neuronal tissues and plays a crucial role in the developing brain. It belongs to a unique class of zinc finger proteins that are essential for the development of the mammalian central nervous system. Interestingly, scientists have discovered that forcing the expression of this gene in combination with other key transcription factors can convert ordinary cells into functional neurons. This has exciting implications for potential future therapies. Mutations in the MYT1L gene have been linked to certain cognitive disabilities and autism spectrum disorder. Multiple variations of the MYT1L protein are produced due to alternative splicing, a process that allows a single gene to code for multiple proteins.

MYT1L is a transcription factor that plays a critical role in neuronal differentiation by specifically shutting down the expression of genes that are not needed for neuron development. This is different from other transcription repressors, which typically only block the development of specific cell types. MYT1L is able to suppress the activity of multiple developmental programs. It also represses the expression of genes that normally hinder the formation of neurons, such as those involved in the Notch signaling pathway, including HES1. Remarkably, MYT1L, along with ASCL1 and POU3F2/BRN2, can transform ordinary cells like fibroblasts into functional neurons in laboratory settings. MYT1L directly attaches to a specific DNA sequence (5'-AAGTT-3') found in the regulatory regions of genes it controls. This binding allows MYT1L to recruit a group of proteins (including SIN3B) to the target gene, effectively silencing its expression. This specific DNA sequence is absent from the regulatory regions of genes involved in neuron development.
