MOV10
Description
The MOV10 (Mov10 RNA helicase) is a protein-coding gene located on chromosome 1.
MOV10 is a putative helicase, an enzyme that in humans is encoded by the MOV10 gene. The stability of the MOV10 protein is controlled by the DCAF12 ubiquitin ligase.
MOV10 is a 5' to 3' RNA helicase involved in a wide range of cellular processes, including mRNA metabolism and translation, modulation of viral infectivity, inhibition of retrotransposition, and regulation of synaptic transmission. It plays a crucial role in innate antiviral immunity by promoting type I interferon production. Mechanistically, MOV10 utilizes IKKepsilon/IKBKE as the mediator kinase for IRF3 activation. MOV10 blocks HIV-1 virus replication at a post-entry step and counteracts HIV-1 Vif-mediated degradation of APOBEC3G through its helicase activity by interfering with the ubiquitin-proteasome pathway. It also inhibits hepatitis B virus (HBV) replication by interacting with HBV RNA, thereby inhibiting the early step of viral reverse transcription. MOV10 contributes to UPF1 mRNA target degradation by translocating along 3' UTRs. It is required for microRNA (miRNA)-mediated gene silencing by the RNA-induced silencing complex (RISC), specifically for both miRNA-mediated translational repression and miRNA-mediated cleavage of complementary mRNAs by RISC. In cooperation with FMR1, MOV10 regulates miRNA-mediated translational repression by AGO2. It restricts retrotransposition of long interspersed element-1 (LINE-1) in cooperation with TUT4 and TUT7, counteracting the RNA chaperone activity of L1RE1. MOV10 facilitates LINE-1 uridylation by TUT4 and TUT7. It is essential for embryonic viability and normal central nervous system development and function. During early brain development, MOV10 plays two critical roles: it suppresses retroelements in the nucleus by directly inhibiting cDNA synthesis, and it regulates cytoskeletal mRNAs to influence neurite outgrowth in the cytosol. MOV10 may function as a messenger ribonucleoprotein (mRNP) clearance factor.
