Description

The TOP1MT (DNA topoisomerase I mitochondrial) is a protein-coding gene located on chromosome 8.

TOP1MT, or DNA topoisomerase I, mitochondrial, plays a crucial role in managing the supercoiling and torsional stress that builds up during mitochondrial DNA replication. It achieves this by temporarily cleaving one strand of the DNA duplex, introducing a single-strand break through a transesterification reaction. This process involves the catalytic tyrosine of TOP1MT attacking the phosphodiester bond, forming a covalent intermediate and releasing a free DNA strand. The free strand then rotates around the intact opposite strand, alleviating the supercoiling. Finally, the DNA strand reattaches to the intermediate, restoring the phosphodiester bond and releasing TOP1MT.
