Description

The TUSC3 (tumor suppressor candidate 3) is a protein-coding gene located on chromosome 8.

TUSC3 is a protein encoded by the TUSC3 gene in humans. This gene is considered a potential tumor suppressor, as it is located within a region frequently deleted in metastatic prostate cancer. TUSC3 is expressed in various human tissues, including prostate, lung, liver, and colon, as well as in many epithelial tumor cell lines. Two distinct protein isoforms of TUSC3 have been identified.

TUSC3 is a component of the N-oligosaccharyl transferase (OST) complex, which attaches a high-mannose oligosaccharide to asparagine residues in newly synthesized proteins. TUSC3 is specifically involved in glycosylating acceptor sites near cysteine residues, often working in conjunction with MAGT1. In its oxidized state, TUSC3 can form temporary disulfide bonds with glycoprotein substrates, facilitating access for the OST complex to the unmodified acceptor site. TUSC3 also has functions within the OST complex independent of its oxidoreductase activity, potentially influencing substrate recognition.
