LIM domain-containing protein 1 is a protein that in humans is encoded by the LIMD1 gene.
LIMD1 is a scaffolding protein involved in various cellular processes including cell fate determination, cytoskeletal organization, gene transcription repression, cell-cell adhesion, differentiation, proliferation, and migration. It positively regulates microRNA-mediated gene silencing and is crucial for P-body formation and integrity. It acts as a hypoxic regulator by facilitating the interaction between prolyl hydroxylases and VHL, leading to efficient HIF1A degradation. It functions as a transcriptional corepressor for SNAI1 and SNAI2/SLUG-dependent repression of E-cadherin transcription. LIMD1 negatively regulates the Hippo signaling pathway, antagonizing YAP1 phosphorylation. It inhibits E2F-mediated transcription, suppressing the expression of genes with E2F1-responsive elements. LIMD1 regulates osteoblast development, function, differentiation, and stress osteoclastogenesis. It enhances TRAF6's ability to activate AP-1 and negatively regulates the canonical Wnt receptor signaling pathway in osteoblasts. LIMD1 may act as a tumor suppressor by inhibiting cell proliferation.
