KSR1


Description

The KSR1 (kinase suppressor of ras 1) is a protein-coding gene located on chromosome 17.

Kinase suppressor of Ras 1 is an enzyme that in humans is encoded by the KSR1 gene.

KSR1 is a component of a multiprotein signaling complex that enhances phosphorylation of Raf family members, ultimately activating downstream MAP kinases. Notably, KSR1 functions as an allosteric activator of BRAF, independent of its kinase activity. Specifically, KSR1 interacts with MAP2K1/MEK1 or MAP2K2/MEK2, leading to dimerization with BRAF and subsequent phosphorylation of MAP2K1/MEK1 and/or MAP2K2/MEK2 by BRAF. Additionally, KSR1 promotes activation of MAPK1 and/or MAPK3 in response to EGF and cAMP. Although some protein kinase activity has been observed in vitro, its physiological relevance remains unclear.
