Description

The IMPACT (impact RWD domain protein) is a protein-coding gene located on chromosome 18.

IMPACT is a protein involved in regulating translation, particularly under stress conditions. It inhibits the activity of the EIF2AK4/GCN2 kinase, preventing the phosphorylation of eIF-2-alpha and ensuring the continued production of proteins. IMPACT also plays a role in neuronal differentiation by stimulating neurite outgrowth.

IMPACT is a translational regulator that maintains high levels of protein synthesis under various stress conditions, such as amino acid starvation, UV-C irradiation, proteasome inhibition, and glucose deprivation. It acts as a negative regulator of EIF2AK4/GCN2 kinase activity, preventing GCN1-mediated activation of EIF2AK4/GCN2, thereby inhibiting eIF-2-alpha phosphorylation and the subsequent downregulation of protein synthesis. IMPACT may also be crucial for regulating translation in specific neuronal cells during amino acid starvation by preventing GCN2 activation and ATF4 synthesis. Its inhibitory action on EIF2AK4/GCN2 contributes to neuronal cell differentiation by promoting neurite outgrowth.
