VAV1
Description
The VAV1 (vav guanine nucleotide exchange factor 1) is a protein-coding gene located on chromosome 19.
VAV1 is a protein encoded by the VAV1 gene, which is classified as a proto-oncogene. It is a member of the Dbl family of guanine nucleotide exchange factors (GEFs) for the Rho family of GTP-binding proteins. VAV1 plays a significant role in hematopoiesis, contributing to the development and activation of T cells and B cells. Interestingly, it has been identified as a specific binding partner for Nef proteins from HIV-1. This interaction triggers morphological changes, cytoskeletal rearrangements, and activation of the JNK/SAPK signaling cascade, ultimately leading to increased viral transcription and replication.
VAV1 couples tyrosine kinase signals with the activation of Rho/Rac GTPases, thereby promoting cell differentiation and/or proliferation.
