Description

The GAS8 (growth arrest specific 8) is a protein-coding gene located on chromosome 16.

GAS8 is a protein that plays a crucial role in regulating cilia function. It is a component of the nexin-dynein regulatory complex (N-DRC), which is essential for ciliary/flagellar motility. GAS8 contributes to the proper alignment and integrity of the distal axoneme and regulates microtubule sliding within motile axonemes. It is also involved in the assembly of the N-DRC linker. GAS8 plays dual roles in both primary (non-motile) and motile cilia. In primary cilia, it regulates Hedgehog signaling, while in motile cilia, it coordinates cilia movement. GAS8 is essential for the proper functioning of motile cilia. It positively regulates the ciliary smoothened (SMO)-dependent Hedgehog (Hh) signaling pathway by facilitating SMO transport into the cilium and promoting SMO activity in a GRK2-dependent manner. GAS8 interacts with various proteins including microtubules, SMO, RAB3B (in its GTP-bound form), DRC1, and DRC7.
