GID4
Description
The GID4 (GID complex subunit 4 homolog) is a protein-coding gene located on chromosome 17.
GID4, also known as Glucose-induced degradation protein 4 homolog or Vacuolar import and degradation protein 24 homolog, acts as a substrate-recognition subunit within the CTLH E3 ubiquitin-protein ligase complex. It selectively receives ubiquitin from UBE2H and facilitates the ubiquitination and subsequent proteasomal degradation of the transcription factor HBP1. GID4 recognizes and binds to proteins and peptides containing a Pro/N-degron, characterized by an unmodified N-terminal proline followed by a small residue. It exhibits the highest affinity for the peptide Pro-Gly-Leu-Trp. Additionally, GID4 binds peptides with an N-terminal sequence of the type Pro-[Ala,Gly]-[Leu,Met,Gln,Ser,Tyr]-[Glu,Gly,His,Ser,Val,Trp,Tyr]. However, it does not bind to peptides with an acetylated N-terminal proline residue.
