Description

The GGA2 (golgi associated, gamma adaptin ear containing, ARF binding protein 2) is a protein-coding gene located on chromosome 16.

GGA2, or Golgi-localized, gamma adaptin ear-containing, ARF-binding protein 2, is a protein encoded by the GGA2 gene in humans. It belongs to the GGA family of coat proteins that regulate protein trafficking between the trans-Golgi network (TGN) and the lysosome. GGA2 contains a VHS domain for sorting mannose 6-phosphate receptors at the TGN and a gamma-adaptin ear domain. It plays a role in cargo molecule regulation and clathrin-coated vesicle assembly. GGA2 interacts with various proteins, including RABEP1, Sortilin 1, BACE2, and CLINT1.

GGA2 plays a crucial role in protein sorting and trafficking between the trans-Golgi network (TGN) and endosomes. It facilitates the ARF-dependent recruitment of clathrin to the TGN and binds ubiquitinated proteins and membrane cargo molecules through a cytosolic acidic cluster-dileucine (DXXLL) motif. GGA2 mediates the export of the GPCR receptor ADRA2B to the cell surface and regulates the retrograde transport of the phosphorylated form of BACE1 from endosomes back to the trans-Golgi network.

GGA2 is also known as VEAR.
