Description

Foxd3 (forkhead box D3) is a transcription factor, a protein that regulates the expression of genes by binding to specific DNA sequences. It plays a crucial role in various biological processes, including embryonic development and organogenesis.

Foxd3 is encoded by the FOXD3 gene located on chromosome 1p36. It belongs to the winged-helix subfamily of transcription factors, characterized by a conserved DNA-binding domain that resembles the wing structure of an insect.

Associated Diseases

Disruptions in Foxd3 function have been linked to several diseases, including:

  • Eyelid malformations: Foxd3 is essential for the proper development of eyelids. Mutations in the FOXD3 gene can lead to eyelid malformations such as blepharophimosis (narrow eyelids) and ptosis (droopy eyelids).
  • Dental anomalies: Foxd3 is involved in the development of teeth. Mutations in the FOXD3 gene can cause dental anomalies such as missing or malformed teeth and crowding.
  • Alopecia areata: Foxd3 plays a role in the regeneration of hair follicles. In alopecia areata, an autoimmune disorder, the immune system attacks hair follicles leading to hair loss. Studies have found that Foxd3 expression is reduced in the hair follicles of alopecia areata patients.
  • Bladder exstrophy: This is a rare congenital birth defect in which the bladder is exposed outside the abdomen. Mutations in the FOXD3 gene have been identified in some cases of bladder exstrophy.
