Description

The FBXO3 (F-box protein 3) is a protein-coding gene located on chromosome 11.

FBXO3 is a substrate recognition component of the SCF (SKP1-CUL1-F-box protein)-type E3 ubiquitin ligase complex, specifically SCF(FBXO3). This complex mediates the ubiquitination and subsequent proteasomal degradation of target proteins, including HIPK2 and potentially EP300. While FBXO3 promotes HIPK2 ubiquitination, the presence of PML prevents its degradation. Interestingly, FBXO3, HIPK2, and PML may work together to activate p53/TP53-dependent transactivation. Moreover, SCF(FBXO3) plays a role in regulating inflammation by ubiquitinating and degrading FBXL2 in response to lipopolysaccharide (LPS). This complex preferentially recognizes FBXL2 phosphorylated at 'Thr-404' and facilitates its ubiquitination.
