EIF3J plays a critical role in protein synthesis by being a subunit of the eukaryotic translation initiation factor 3 (eIF-3) complex. This complex is essential for various steps in protein synthesis initiation, including the association with the 40S ribosome, recruitment of key initiation factors (eIF-1, eIF-1A, eIF-2:GTP:methionyl-tRNAi, and eIF-5) to form the 43S pre-initiation complex (43S PIC), and facilitating mRNA recruitment to the 43S PIC for scanning and AUG recognition. Additionally, eIF-3 is involved in the disassembly and recycling of post-termination ribosomal complexes, preventing premature joining of ribosomal subunits before initiation. Furthermore, eIF-3 selectively targets and initiates the translation of mRNAs involved in cell proliferation, including those related to cell cycle, differentiation, and apoptosis, through distinct RNA stem-loop binding mechanisms that can either activate or repress translation.
