Description
The DNPH1 (2'-deoxynucleoside 5'-phosphate N-hydrolase 1) is a protein-coding gene located on chromosome 6.
DNPH1 is an enzyme involved in a nucleotide salvage pathway that removes epigenetically modified 5-hydroxymethyl-dCMP (hmdCMP). This process occurs in two steps: deamination to create cytotoxic 5-hydroxymethyl-dUMP (hmdUMP), followed by hydrolysis of hmdUMP into 5-hydroxymethyluracil (hmU) and 2-deoxy-D-ribose 5-phosphate (deoxyribosephosphate). DNPH1 specifically catalyzes the second step, breaking the N-glycosidic bond in hmdUMP. This degradation prevents the integration of this cytotoxic nucleotide into the genome.
